Supplier: Biotium

5(6)-FAM SE (full name: 5-(and-6)-Carboxyfluorescein, succinimidyl ester mixed isomers) is an amine-reactive green fluorescent dye widely used for labeling proteins or other molecules that contain a primary or secondary aliphatic amine. The coupling reaction is usually carried out at pH 8-9.5. The amide linkage from the coupling reaction is much more stable than the thiourea linkage formed from the coupling of an amine and an isothiocyanate.

ADAM Substrate

Supplier: ENZO LIFE SCIENCES

Fluorogenic substrate for ADAM8 (kcat/Km = 1,0×105 M-1s-1), ADAM10 (6,2×103), ADAM12 (2,8×105), TACE (TNF-α converting enzyme, ADAM17) (4,3×105), and likely other ADAMs, especially those that cleave TNF-α type sequences. Also efficiently cleaved by MMP-1, MMP-2, MMP-3, MMP-8, MMP-9, MMP-13, and MMP-14. FAM fluorescence is quenched by the Dabcyl group until cleavage separates them. Ex.: 485 nm, Em.: 530 nm. This substrate is useful for inhibitor screening and kinetic analysis.

TACE substrate II

Supplier: ENZO LIFE SCIENCES

Sensitive fluorogenic substrate for TACE (TNFalpha converting enzyme, ADAM17). Also cleaved by ADAM10 and likely other ADAMs and MMPs. The kcat/Km of TACE for this substrate is 7-fold higher than for the same peptide with the Mca-Dpa fluorophore-quencher pair. FAM fluorescence is quenched by the TAMRA group until cleavage separates them. Ex.: 485 nm, Em.: 535 nm, although the following Ex/Em can also be used: 490,494/515,520 nm. This substrate is useful for inhibitor screening, kinetic analysis, and cellular activity assay.

5(6)-Carboxyrhodamine 110 succinimidyl ester ≥85% (by HPLC) amine-reactive fluorescent dye

Supplier: Biotium

5-(and-6)-Carboxyrhodamine 110, SE (5-(and-6)-Carboxyrhodamine 110, succinimidyl ester, mixed isomers), also known as Carboxyrhodamine 110-NHS, is a superior green fluorescent dye that readily reacts with primary or secondary amines. For many applications, the dye is preferred over 5-(and-6)-carboxyfluorescein succinimidyl ester (5(6)-FAM) or FITC because of its exceptional photostability and fluorescence insensitivity to pH (4-9).
